Background
ALKBH5 is a nuclear 45-kDa Fe(II)- and 2-oxoglutarate-dependent dioxygenase that selectively demethylates N6-methyladenosine (m6A) in single-stranded RNA, thereby dynamically modulating mRNA splicing, export, stability, and translation; highly expressed in testes and within nuclear speckles, it is essential for spermatogenesis and fertility, while in somatic tissues it orchestrates glucose and lipid homeostasis through independent regulation of GCGR and mTORC1 signaling, and its dysregulation—via hypoxia-inducible upregulation or cancer-associated overexpression—drives stemness, proliferation, and chemoresistance in breast cancer, glioblastoma, and ovarian cancer by demethylating oncogenic transcripts such as NANOG and FOXM1, positioning ALKBH5 as a central epitranscriptomic hub linking metabolism, reproduction, and malignancy.
