Background

Multi Nuclease also called broad-spectrum nucleic acid enzyme, is a kind of comes from Serratia Marcescens restriction endonuclease. It is capable of degradation of all forms of DNA and RNA (double-stranded, single-stranded, linear, circular or super helical forms) under a very wide range of conditions (6Murea, 0.1M Guanidine, 0.4%TritonX100, 0.1%SDS, 1mM EDTA, 1mM PMSF). The expression and purification of this product in Escherichia coli(E.coli) through genetic engineering can not only reduce the viscosity of cell supernatant and cell lysate in scientific research, but also improve the efficiency of protein purification and functional research. It can also be used in virus purification, vaccine production, protein and polysaccharide pharmaceutical industry as a host residual nucleic acid removal reagent, reducing the host residual nucleic acid to the peak (pg) level to improve the efficacy and safety of biological products. And can effectively prevent human peripheral blood monocyte (PBMC) clumping in cell therapy and vaccine research.

Components

Storage Solution: ≥250U/μl Ultra Nuclease (Tag free)、10mM Tris (pH7.4), 500mM NaCl, 2mM MgCl2, 50% glycerol

Protocol

1. Sample preparation:

Adherent cells: Remove the medium, clean the cells with PBS, and remove the supernatant.

Suspension cells: Cells were collected by centrifugation, cleaned with PBS, centrifuged at 6,000rpm for 10min, and precipitates were collected.

Escherichia coli: The bacteria were collected by centrifugation, cleaned once with PBS, centrifuged at 8,000rpm for 5min, and precipitates were collected.

2. Sample treatment:

The collected cell precipitates are cleaved according to the ratio of mass (g) to volume (mL) to 1: (10~20). Cells can also be cleaved mechanically or chemically on ice or at room temperature (1g cells are about 109).

3. Enzyme addition:

the proportion of 1g cell precipitation digested by 250Units is required. You can also choose the addition plan according to the recommended dosage in the table above, increase the amount of enzyme within a certain range, and reduce the digestion time accordingly.

4. Supernatant acquisition:

The supernatant of cell lysis solution was obtained by centrifugation at 12,000rpm for 30min, and then subsequent related experiments were conducted.

Very Disappointed with this Vacuum
After having a Shark Robot Vacuum for many years, my husband decided to upgrade to newer technology (easier boundary settings, better mapping etc) On a good note, when this thing works - it works great. One of my biggest complaints with the Shark was I had to put physical boundary strips at the top of the stairs etc. This Eufy is very easy to set "no go zones" on the app, works famously in that aspect. This vac also seems to "work" in a more organized manner. I can see on the app exactly what was cleaned after every scheduled cleaning etc. The Shark would just buzz around the house all willy nilly, but this Eufy seems to be very organized in the cleaning, not missing anywhere. Now for the most aggravating part. I have my Eufy scheduled to clean every night @ 1:00 am, while we are sleeping. Most nights @ 1:15 am I get a notification on my phone "Roller Brush Stuck". Well obviously I don't see the notification until morning. I go "find" Eufy (which is easy with the app - shows you right where she is - another nice thing compared to the Shark) Pull the Roller Brush, which mind you, nothing is wrong with, it is not stuck, there is nothing wrapped around it etc. Put it back in & restart. This has done this since day one, brand new out of the box. After doing some scouring of the Internet, I see that this is a big complaint with many other owners of this model. From what I gather, Eufy is aware of this and it is a known problem. I called Eufy Customer Care after about using this for one week. The phone call lasted over 45 minutes (part of my life that I will never get back!) Now mind you, the representative was very helpful & thorough. We walked through a couple procedures, (The Rep had me pull the roller brush etc & report to her what I was "seeing") Which again, I had performed everything the Rep had me do religiously for the past week, but No, she like wouldn't take my word for it, I had to do all this again while on the phone with her - - almost like she didn't believe me at first. Next, the Rep offered to send me a "newer design roller brush" Ok, cool, if it solves the problem let's do it - I just wanted to get off the phone & on with my day. Oh wait, there are no new design roller brushes in stock. Now I'm told to send just the robot back (not the dock) I told the Rep to send me a call tag, as I wasn't going to pay shipping on a "brand new used one week" item. Oh wait, now she has located the replacement roller brushes. So here we are back to the original solution, she will send me a replacement roller brush. Took my name, my address, phone number, name of first & last born children etc. Just a long drawn out process to send me the "fix" for their known manufacture problem. So to make my long story even longer, here I sit a month and a 1/2 later without ever receiving the replacement brush and an unreliable product who doesn't complete her "mission" most nights. I wish this vacuum was still within the 30 day Amazon return period, because I would have sent it back after experiencing this defect. However my husband had bought this for me for Christmas (before the date that Amazon offers their extended holiday returns window) I didn't even unbox this vacuum before the 30 days were up. The App with the mapping & the expansive scheduling options is great compared to the Shark, but I bought the Eufy to vacuum, which she fails @ nightly.

Absolutely amazing! Working flawlessly for over a month, so far.
Color: White
I am very pleased with the performance so far. The strengths are the ease of setup, the accuracy of its mapping and carpet/floor recognition, the quality of its cleaning and mopping, and the entertainment of just watching. There are weaknesses. The documentation is sparse and pretty useless and tech support has been ghosting me..no responses. So hunt and peck with the App, mostly. Hard to really customize by trial and error. While it vacuums very well, it does not empty as well, really requiring the owner to manually empty the dust bin. No big deal. Maintenance recommendations are hidden on the website and I suspect it is advisable to do some maintenance after every use. The strengths win, however.

User friendly, Pet Hair Approved, Quite Powerful Robot Vacuum
Color: White
We prviously had an Eureka E10 vacuum and mop robot for 4 years. We had no idea what we were missing until we purchased this Narwal Freo X10 Pro. It maps very fast. It is extremely quite. The vacuum then mop with a refreshed mop head solution is much better than dragging around a robot mop pad with no way to clean it without removing and washing it. We have 3 Australian Sheppards that shed alot. This was the number one concern, whereas the prvious unit needed daily attention. Not a single hair tangle in the floating rotating brush of this unique Narwal Freo X10 Pro Robot vacuum. 4 months of no robot management is wonderful. The application was easy to use with a ton of customizable options. The AI option to recognize various floor types and throw rugs ensures a complete clean and quick simple setup. Lastly the price vs product was very reasonable. Great product! After spending many months researching, we are glad we made this investment.

After five months, it's basically a brick
Color: White
We've had this robot for five months, and it isn't durable. It worked fine for a couple months. But now it works fine for maybe 20 minutes and then just starts spinning around or going back in forth in the same area wasting its battery. The endless troubleshooting does not help. Cleaning all the sensors, resetting the robot, remapping the house ... Nothing helps. It is only being used in about 1100 sq ft of total space, so it's just not that complicated. It's been trying (and failing) to vacuum our smallish living room for nearly five hours, after several resets/etc. Back when it did function, the vacuum is rather loud if you want it to actually pick up anything big enough to see. The vacuum and mop are both terrible at inner corners, and the intersection between rugs and hardwood flooring is a total miss. There's like a 12" strip along all rugs where mopping never occurs. Definitely not a machine to be used without supplemental sweeping and mopping. The machine insists on starting rugs at the corner, so it pulls up even pretty heavy rugs sometimes. It's sucks up items like socks instead of avoiding them. Also, the machine has never successfully left alone no-vacuum rugs or no-go areas, so some rooms (like with high pile rugs or Christmas tree stands) just don't work without physical barricades. Really, really makes me miss the old reliable roombas...

This vacuum is a game changer
Color: White
Not all robotic vacuums are the same, and after testing so many of them in my life this one has outperformed the others hands down! From the suction power, to the battery life, and then the app I can't find a single thing I don't love. The robot is and to pickup small crumbs, dirt, the typical messes, but most importantly it picks up kitty litter like it's nothing. This is an instant game changer. I no longer need to clean up after a robot. It's amazing! The battery life is great! I have a 2500 sq ft house and it can handle all of it before charging. The "home base" makes this effortless where I just schedule my cleans and it takes care of the rest. The app is really nicely built. It's simple and easy to navigate but it also gives me all of the advanced options such as invisible walls and no-go zones. Overall, this is robotic vacuum is my new go-to and I look forward to using it for many years to come!
